Functional complexes of the bacterial ribosome undergo large conformational changes during the course of translating mRNA into proteins. Recent dramatic progress in the determination of ribosome structure has provided some of the best evidence for such changes. Using both ensemble and single molecule approaches we are conducting detailed kinetic studies in order to elucidate both how specific structural changes are pertinent for specific steps of the translation cycle and how sequences within mRNA modulate the rate of translation by E. coli ribosomes.
